LIFECODES Donor Specific Antibody

Donor Specific Antibodies (DSA) have been shown to be involved in both chronic and acute rejection of transplanted allografts.  Gen-Probe provides solutions for the detection of DSA in transplant patients.  Two product options are available providing a choice in testing platforms.

LIFECODES DSA Donor Specific Antibody Detection

DSA makes it possible to screen for the presence of antibodies directed against the HLA antigens of the donor. It is ideal for pre- and post-transplant monitoring.

Donor lymphocytes are first isolated and lysed. The lysate is then incubated with Class I specific and Class II specific beads. The HLA antigens of the donor are bound to the specific beads via capture monoclonals. The beads are then incubated with the serum and any HLA antibodies against the donor are detected using the Luminex analyzer after addition of anti-human IgG conjugated with PE.

LIFECODES MicroAMS

MicroAMS is designed to detect donor-specific HLA alloantibodies in transplant recipients that may lead to early graft loss or chronic rejection. Class I and Class II glycoproteins are solubilized from donor cells or tissue and captured using class-specific monoclonal antibodies which have been immobilized in the microwells. The recipient serum of interest is then added. A color change indicates the presence of donor-reactive antibodies.1

  • No interference from antibodies to non-HLA glycoproteins
  • Uses lysates (solubilized donor glycoproteins) that can be made from lymphocytes isolated from peripheral blood or cells from spleen
  • Reduced volume requirement for both donor lysate and patient serum
  • Lysates can be prepared in advance and stored for an extended period at -80°C
