Nucleic Acid Extraction

We have extensive experience in providing research and regulatory compliant nucleic acid extraction and purification services. Our Manchester-based laboratory facilities are BS:EN ISO 9001:2000 and CLIA accredited. Our service provides extraction to the worlds leading medical, pharmaceutical and clinical research organisations and as a result we have expertise in working with a wide range of sample types submitted from across the globe. DNA and RNA prepared by our service is suitable for all downstream applications including sequencing and genotyping. We also routinely generate archival quality nucleic acid samples. A Project Manager (PM) will be assigned to act as the sole point of contact for all projects. The PM will be responsible for all aspects of the project lifecycle including organisation of sample shipments, scheduling, processing & return of data.

DNA Extraction
We provide a rapid and affordable service which generates high quality and high yield DNA using our proprietary Nucleon product chemistry. Using validated procedures we are able to extract DNA from a broad range of biological matrices including but not limited to:

  • Whole blood
  • Lymphocytes, buffy coats, plasma and sera
  • Saliva collected into commercially available kits for e.g. Oragene DNA Kits
  • Frozen tissue and biopsy material
  • Formalin Fixed Paraffin Embedded Tissue (FFPE)
  • Buccal scrapes / swabs and mouthwash samples
  • Cultured cells (mammalian and bacterial)
  • Blood cards

Rapid DNA Extraction Service
We now offer a high throughput DNA extraction system capable of extracting and purifying up to 250µL human whole blood. Samples are processed using a commercially available magnetic bead-based chemistry. The resultant DNA is suitable for use in a range of downstream applications including SNP genotyping on Illumina and Affymetrix platforms and Taqman/qPCR SNP assays, Sequencing and Copy Number Variation.

RNA Extraction
In addition to total RNA isolation we also offer a miRNA isolation service. To obtain high quality RNA we recommend that samples should be immediately cryo-preserved following collection or transferred to an RNA stabilisation fluid. We have proven experience in isolating RNA using TRIzol methodology and commercially available kits from a number of different sample types including but not limited to:

  • Whole blood (including samples collected into Paxgene tubes)
  • Formalin Fixed Paraffin Embedded Tissue (FFPE)
  • Frozen tissue and biopsy material
  • Cultured cells (mammalian and bacterial)

Post Extraction Services
Purified DNA and RNA can be quantitated by either optical density at A260nm and A280nm wavelengths. We also offer Picogreen measurement for DNA samples and Ribogreen for RNA samples. Nucleic acid quality can also be assessed using agarose gel electrophoresis. We have the ability to assess RNA quality on an Agilent 2100 Bioanalyzer. Nucleic acids can be normalised to a concentration and volume of your choice and reformatted in a variety of output plate and tube types, for e.g. PCR plates, 2D-barcoded tubes. Upon completion of projects, we provide a detailed quality report to suit individual client requirements.
